This element represents a locus control region (LCR) that can confer T cell-specific, copy number-dependent, position-independent expression on the CD2 gene in transgenic mice. It is located immediately downstream of the 3' terminal exon of the CD2 gene, and is characterized by three DNase I hypersensitive sites and a core enhancer region. This LCR and the enhancer sub-region have been used to drive T cell-specific expression of several heterologous genes in various vector systems, including retroviral and lentiviral vectors. A truncated version of the LCR has been shown to be heterochromatin-sensitive, and has thus been useful for studying chromatin-related processes, such as triplet-repeat-associated position effect variegation.
